3-methyl-10-ethyl-deazaflavin is an enhanced, highly brain permeable form of 5-deazaflavin, an analog of vitamin B2 shown to directly augment electron transport and mitochondrial energy production. Investigated for its potential role in neuronal energy metabolism, mitochondrial function, and cognitive-health research, preclinical evidence suggests that it may have the following effects:

  • Increases neuronal ATP output by directly facilitating electron transport in neuronal mitochondria.
  • May help neurons tolerate high-energy stress in experimental models involving calcium overload and intense depolarization.
  • May slow cognitive-aging in research on impaired neuronal energy metabolism.

3-methyl-10-ethyl deazaflavin belongs to the deazaflavin family and has been described as a self-redox-capable molecule with activity resembling cellular redox cofactors like vitamin B2. Its unique structural alterations vastly improve its membrane permeability, allowing it to cross the BBB and enter neuronal mitochondria where it has been shown to directly increase ATP output.

Research by the Medical University of Tokyo on pretreatment with TND1128 reduced stress-induced Ca2+ elevation in both the cytosol and mitochondria, suggesting a possible role in helping neurons maintain energy-dependent calcium regulation under demanding conditions. Uncontrolled fluctuation of Ca2+ in neurons has been established as a primary driver of brain aging and excitotoxicity. This makes the molecule especially interesting for research into mitochondrial support, neuronal stress tolerance, cognitive aging, and cellular models of neurodegenerative vulnerability.

The same modifications that give 3-methyl-10-ethyl deazaflavin its outstanding membrane penetration also limit its oral bioavailability in dry powder form. This is because lipophilic (fat-preferring) molecules disperse very poorly in gastrointestinal fluid. Because gut fluid is largely aqueous, hydrophobic powders tend to clump together tightly and sail through the GI tract with minimal absorption.

Mitotek's nanoemulsion delivery system is critical to making this potentially powerful molecule bioavailable for oral administration. Nanoemulsification disperses the compound into very small oil-phase droplets, helping create a more practical gut-compatible delivery that spreads the molecule across the gut for maximal absorption.
